Een nieuw bioengineered fragmentpeptide van Vasostatin-1 oefent farmacologische activiteiten van gladde spieren en anti-angiogene effecten uit through het blokkeren van de VEGFR-signaleringsroute
Chromogranine A (CgA) is een hydrofiel glycoproteïne dat wordt afgegeven door postganglionaire sympathische neuronen. CgA bestaat uit een enkele peptideketen die talrijke gepaarde basische residuen bevat, die typische splitsingsplaatsen in prohormonen zijn om bioactieve peptiden te genereren. Het wordt erkend als een diagnostische en prognostische serummarker voor neuro-endocriene tumoren. Vasostatine-1 is een van de meest geconserveerde regio’s van CgA en heeft numerous remmende biologische activiteiten. In deze studie werd een nieuw peptidefragment gesynthetiseerd dat drie typische functionele structuren van Vasostatin-1 bevat. Dit unieke bioengineered Vasostatin-1 Derived Peptide (genaamd V1DP) omvat een sterk geconserveerd domein tussen gewervelde soorten in het N-terminale gebied, bestaande uit een disulfidebrug gevormd door twee cysteïneresiduen op respectievelijk aminozuurposities 17 en 38. Trouwens, V1DP bevat twee significante tripeptide-herkenningssequenties: de aminozuurtriplets, RGD en KGD. Onze gegevens toonden aan dat V1DP een dosisafhankelijke relaxatie van de arteriële gladde spier van de rat kon induceren en ook de contractie-activiteit van de gladde spier van de baarmoeder van de rat kon verhogen. Wat nog belangrijker is, we ontdekten dat V1DP de proliferatie van kankercellen remt, de HUVEC-celmigratie moduleert en een anti-angiogenese-effect vertoont, zowel in vitro en in vivo.
We hebben het feitelijke mechanisme van V1DP verder onderzocht en onze resultaten bevestigden dat V1DP de vasculaire endotheliale groeifactorreceptor (VEGFR) -signalering remt. We koppelden V1DP aan de apo-structuren van VEGFR2 en onderzochten de stabiliteit van het peptide in de eiwitpockets. Onze simulatie- en vrije-energieberekeningen gaven aan dat V1DP kan binden aan het katalytische domein en regulatoire domeinzakken, afhankelijk van of de conformationele toestand van het eiwit JM-in of JM-out is. Al met al suggereerden onze gegevens dat V1DP een rol speelt als regulator van de endotheelcelfunctie en farmacologische homeostase van gladde spieren. V1DP is een in water oplosbaar en biologisch stabiel peptide en zou zich verder kunnen ontwikkelen als een anti-angiogeen geneesmiddel voor de behandeling van kanker.

Ontwikkeling van een peptide dat DNA-herstel remt door de binding van Artemis en DNA Ligase IV te blokkeren om de radiogevoeligheid van tumoren te verbeteren
Doel: Artemis en DNA-ligase IV zijn twee cruciale elementen in de NHEJ-route van DNA-herstel, die respectievelijk als nuclease en DNA-ligase fungeren. Verbeterde cellulaire stralingsgevoeligheid door remming van beide eiwitten draagt bij aan een veelbelovende benadering om moleculaire gerichte radiosensitizers te ontwikkelen. De interactie tussen Artemis en DNA Ligase IV is vereist voor de activering van Artemis als nuclease bij 3′-overhangend DNA; daarom willen we een remmend peptide genereren dat gericht is op de interactie tussen Artemis en DNA Ligase IV voor de ontwikkeling van nieuwe radiosensitizers.
Methoden: We hebben het peptide BAL gesynthetiseerd dat bestaat uit de interactieresiduen van Artemis met DNA-ligase IV. Het radiosensibilisatie-effect van BAL werd geëvalueerd door middel van een kolonievormingsassay. De effecten van BAL op door bestraling geïnduceerd DNA-herstel werden geëvalueerd met Western Blotting en immunofluorescentie. Effecten van BAL op celproliferatie, stopzetting van de celcyclus en celapoptose werden beoordeeld through CCK-Eight en flowcytometrie-assays. De mogelijke synergetische effecten van BAL en bestraling in vivo werden onderzocht in een xenograft muismodel.
Resultaten: Resultaten tonen aan dat het gegenereerde peptide BAL dat de interactie tussen Artemis en DNA Ligase IV blokkeert, de stralingsgevoeligheid van GBC-SD- en Hela-cellijnen important verhoogt. BAL verlengt DNA-herstel na bestraling; BAL en bestraling vertonen synergetische effecten op celproliferatie, celcyclus en celapoptose; en deze functies zijn allemaal gerelateerd aan DNA-ligase IV. Ten slotte hebben we het endogene radiosensibilisatie-effect van BAL in een xenograft-muismodel bevestigd.
Conclusies: Samenvattend, het remmende peptide BAL dat zich richt op Artemis en DNA Ligase IV-binding functioneert met succes als een nieuwe radiosensitizer die DNA-herstel vertraagt en synergiseert met bestraling om celproliferatie te remmen, celcyclusstilstand te induceren en celapoptose te bevorderen.
Een oraal beschikbare PD-1/PD-L1-blokkerende peptide OPBP-1-geladen trimethylchitosan-hydrogel voor immunotherapie van kanker
Blokkade van het immuuncontrolepunt PD-1/PD-L1 met monoklonale antilichamen toonde ongekende klinische werkzaamheid bij veel kankers. Maar de oraal beschikbare remmers met een laag molecuulgewicht blijven in de kinderschoenen. In vergelijking met kleine moleculen vertoont peptide een betere selectiviteit en minder bijwerkingen, maar een slechte halfwaardetijd en een grote uitdaging om oraal te worden toegediend. Hier hebben we een proteolyse-resistente D-peptide OPBP-1 (Oral PD-L1 Binding Peptide 1) ontwikkeld die selectief PD-L1 kan binden, PD-1/PD-L1-interactie important kan blokkeren en IFN-γ (interferon γ) secretie kan verbeteren van CD8 + T-cellen in menselijke PBMC’s (perifere mononucleaire bloedcellen). OPBP-1 zou de tumorgroei in muriene colorectale CT26- en melanoom B16-OVA-modellen important kunnen remmen bij een relatief lage dosis van 0,5 mg/kg, met verbetering van de infiltratie en functie van CD8 + T-cellen. Interessanter is dat orale afgifte van OPBP-1 geladen TMC (N, N, N-trimethylchitosan) hydrogel ( OPBP-1@TMC) vertoonde een zeer goede orale biologische beschikbaarheid van OPBP-1 (52,8%) en verlengde halfwaardetijd (14,55 uur) bij ratten, en remde ook important de tumorgroei in het CT26-model. Concluderend hebben we een PD-1/PD-L1-blokkerend peptide OPBP-1 ontdekt en geoptimaliseerd, through en vervolgens geladen in een op TMC gebaseerd hydrogel oraal toedieningssysteem, om de orale biologische beschikbaarheid van het peptidegeneesmiddel maximaal te verhogen en de tumorgroei effectief te remmen. . Deze resultaten openden een nieuw perspectief voor de ontwikkeling van orale geneesmiddelen bij immunotherapie van kanker.
Intermoleculaire interactieanalyses op SARS-CoV-2 Spike Protein Receptor Binding Area en Human Angiotensine-Changing Enzyme 2 Receptor-blocking Antibody/Peptide met behulp van fragmentmoleculaire orbitale berekening
Het spike-glycoproteïne (S-proteïne) bemiddelt SARS-CoV-2 binnenkomst through intermoleculaire interactie met humaan angiotensine-converterend enzym 2. Het receptorbindende domein (RBD) van het S-proteïne wordt als cruciaal beschouwd voor deze interactie en fungeert als het doelwit van talrijke neutraliserende antilichamen en antivirale peptiden. Deze studie gebruikte de fragment moleculaire orbitale methode om de interacties tussen de RBD en antilichamen/peptiden te analyseren en cruciale residuen te extraheren die als epitopen kunnen worden gebruikt. De interacties geëvalueerd als interfragment interactie energiewaarden tussen de RBD en 12 antilichamen / peptiden toonden een redelijk goede correlatie met de experimentele activiteit pICso 50 ( R 2 = 0,540). Negen residuen (T415, Ok417, Y421, F456, A475, F486, N487, N501 en Y505) werden bevestigd als cruciaal. Analyses van de ontleding van de energie van een paar interacties toonden aan dat waterstofbruggen, elektrostatische interacties en π-orbitale interacties belangrijk zijn. Onze resultaten bieden essentiële informatie voor het begrijpen van SARS-CoV-2-antilichaam / peptide-binding en kunnen een rol spelen in het toekomstige ontwerp van antilichaam / antivirale geneesmiddelen.
